(MT1-MMP) implicates the complex between MT1-MMP and tissue inhibitor of MMP-2 (TIMP-2) as a receptor for proMMP-2. To dissect this model and assess the pathologic significance of MMP-2 activation, an artificial receptor for proMMP-2 was created by replacing the signal sequence of TIMP-2 with cytoplasmic/transmembrane domain of type II transmembrane mosaic serine protease (MSP-T2). Unlike TIMP-2, MSP-T2 served as a receptor for proMMP-2 without inhibiting MT1-MMP, and generated TIMP-2-free active MMP-2 even at a low level of MT1-MMP. Thus, MSP-T2 did not affect direct cleavage of the substrate testican-1 by MT1-MMP, whereas TIMP-2 inhibited it even at the level that stimulates proMMP-2 processing. Expression of MSP-T2 in HT1080 cells enhanced MMP-2 activation by endogenous MT1-MMP and caused intensive hydrolysis of collagen gel. Expression of MSP-T2 in U87 glioma cells, which express a trace level of endogenous MT1-MMP, induced MMP-2 activation and enhanced cell-associated protease activity, activation of extracellular signal-regulated kinase, and metastatic ability into chick embryonic liver and lung. MT1-MMP can exert both maximum MMP-2 activation and direct cleavage of substrates with MSP-T2, which cannot be achieved with TIMP-2. These results suggest that MMP-2 activation by MT1-MMP potentially amplifies protease activity, and combination with direct cleavage of substrate causes effective tissue degradation and enhances tumor invasion and metastasis, which highlights the complex role of TIMP-2. MSP-T2 is a unique tool to analyze physiologic and pathologic roles of MMP-2 and MT1-MMP in comparison with TIMP-2. ©2008 American Association for Cancer Research.全文公開200911

metabolism of some clinical drugs, carcinogens, and steroid hormones. The molecular mechanisms of the inducible expression of UGT2B7 in response to xenobiotics have not been fully clarified. We sought to investigate whether the UGT2B7 is under the control of NF-E2 p45-related factor 2 (Nrf2), a key transcriptional factor regulating the expression of cytoprotective enzymes. METHODS: HepG2, HuH7, HLE, and Caco-2 cells were treated with sulforaphane (SFN), and the UGT2B7 mRNA levels were determined by real-time reverse transcriptase PCR. These cells were genotyped for the UGT2B7*2 (H268Y) allele using the PCR-restriction fragment length polymorphism method. Luciferase analyses and gel shift analyses were performed to identify the responsive regions for Nrf2 signaling. RESULTS: The UGT2B7 mRNA was induced by SFN in HepG2 and HuH7 genotyped as UGT2B7*1/*1, but not in HLE and Caco-2 cells genotyped as UGT2B7*2/*2. In HepG2 cells, the UGT2B7 protein level and morphine glucuronosyltransferase activity were also significantly induced by SFN. The induction was prominently decreased with small interfering RNA for Nrf2. In the 5'-flanking region (-2.5 kb) of the UGT2B7*2 allele, a 324-base pair insertion at -2067 and 12 single nucleotide polymorphisms simultaneously existed. Luciferase analyses and gel shift analyses revealed that an antioxidant responsive element at -1170 was responsible for the transactivation by Nrf2. In addition, a region from -990 to -858 on the UGT2B7*1 allele was also responsible for the transactivation by Nrf2. Abrogation of the Nrf2-dependent transactivation of the UGT2B7*2 allele was owing to the single nucleotide polymorphism -900A>G. CONCLUSION: UGT2B7 is transcriptionally regulated by Nrf2, but the mechanism is hindered by polymorphisms in the promoter region of UGT2B7*2. The allele-specific mechanism may cause variability of the glucuronidation in response to oxidative stress.全文公開200908

The cyclic pentavanadate units sandwich the edge-sharing octahedral dimanganese core through, coordination of the oxido group of the pentavanadate. A. dinuclear cobalt complex with, a cyclic decavanadate, [Co 2(OH2)2(VO3)10]6- (2), was also synthesized. The structure analysis reveals a dinuclear cobalt complex with a macrocyclic decavanadate, which is composed of 10 VO4 units joined by the vertex. Sharings. The CoO6 octahedrons are edge-shared, with each cobalt octahedron coordinated to five oxido groups from the decavanadate. The remaining site is occupied by - water. The coordinated water molecules are supported with hydrogen bonds in two directions. Complex 2 in acetonitrile shows no reactivity with dioxygen even at low temperature, and the cyclic voltammogram of 2 shows no redox chemistry in acetonitrile. Complex 2 exhibits chromism by water exposure both, in the solid state and. in acetonitrile. Complex 2 is green-yellow in color, and. the addition of water causes the complex to turn brown. After heating the sample, it returns to its original color in a reversible manner. The EXAFS data in acetonitrile is also reported, and is consistent with the solid-state structure. © Wiley-VCH Verlag GmbH & Co. KGaA.

Peripheral, as well as intracerebroventricular, administration of nesfatin-1 suppresses feeding behavior, though underlying mechanisms are unknown. In this study, we examined effects of nesfatin-1 on cytosolic Ca2+ concentration ([Ca2+]i) in the neurons isolated from the vagal afferent nodose ganglion of mice. Nesfatin-1 at 10-10-10-8 M increased [Ca2+]i in the isolated neurons in a concentration-dependent manner, and at 10-8 M it increased [Ca2+]i in 33 out of 263 (12.5%) neurons. These responses were inhibited under Ca2+-free conditions and by N-type Ca2+ channel blocker, ω-conotoxin GVIA. All the nesfatin-1-responsive neurons also exhibited [Ca2+]i responses to capsaicin and cholecystokinin-8. These results provide direct evidence that nesfatin-1 activates vagal afferent neurons by stimulating Ca2+ influx through N-type channels, demonstrating the machinery through which peripheral nesfatin-1 can convey signals to the brain. © 2009 Elsevier Inc. gulation of many phase II detoxifying enzymes and oxidative-stress-inducible genes in many tissues including bone and cartilage. However, little attention has been paid to precise role of nrf2 on osteoblasts and chondrocytes. In the present study, we therefore investigated the role of nrf2 in the regulation of cell proliferation, differentiation and maturation using both MC3T3-E1 and ATDC5 cells. In MC3T3-E1 cells stably transfected with nrf2 (MC3T3-E1-nrf2 clone), the differentiation-dependent induction of alkaline phosphatase activity was significantly inhibited in addition to decrease of the mineralized matrix formation. Moreover, expression of mRNAs for osteocalcin and type I collagen was also markedly attenuated in MC3T3E1-nrf2 clone but not in MC3T3E1-empty vector clone. These inhibitory effect on cell differentiation was also observed in ATDC5 cells stably transfected with nrf2. Moreover, nrf2 significantly impaired the runx2-dependent enhancement of osteocalcin promoter activity. These data suggest that nrf2 may negatively regulate the differentiation of both osteoblasts and chondrocytes through inhibition of the runx2 dependent transcriptional activity.

We have also carried out a structure-activity relationship (SAR) study of 3-(indol-2-yl)quinoxalin-2-ones, which showed a potent inhibitory activity toward the vascular endothelial growth factor (VEGF)-induced proliferation of human mesangial cells and the VEGF-induced auto-phosphorylation of human umbilical vein endothelial cells.2) Moreover, one of these compounds has a potent medicinal effect based on anti-angiogenic action, by oral administration2) (Chart 1, 9). However, since the existing synthetic methods1) for the preparation of 3-(indol-2-yl)quinoxalin-2-ones consist of multiple steps some of which require strict anhydrous conditions, a convenient and simple synthetic method in place of the existing method is desirable. As a result of the investigations into the synthetic procedures, 3-(3-substituted indol-2-yl)quinoxalin-2-ones can be easily prepared by the condensation of 3-substituted indoles with quinoxalin-2-ones in the presence of trifluoroacetic acid (TFA). Herein, we report the examination of these reaction conditions and the application of this new synthetic method to the synthesis of the derivatives as VEGF inhibitors. © 2007 Pharmaceutical Society of Japan.

iamagnetic and paramagnetic compounds and in crystals having a modulated structure. The spectra and T1 of NMR in solids were measured for [M(H2O)6][AB6] crystals to investigate the static and dynamic structure of H2O and [M(H2O)6]2+ as well as the dynamics of the electron spin in the paramagnetic M2+ ion. The physical properties of [M(H2O)6] [AB6] were found to be as follows: (1) The spin-lattice relaxations of the electron spin of M2+ in [M(H2O)6] [SiF6] (M2+ = Fe 2+ , Co2+ , Ni2+) are dominated by the Orbach process, the Orbach process, and the Raman process, respectively. The spin-lattice relaxation of the electron spin of Cu2+ in [Cu(H 2O)6][PtCl6] is caused by jumping between the Jahn-Teller configurations. (2) In [Cu(H2O)6] [PtCl 6], H2O and [Cu(H2O)6]2+ undergo 180° flips and jumping between the different Jahn-Teller configurations, respectively. A weakening of the hydrogen bond O-H⋯Cl upon deuteration results in a lowering of the transition temperature. (3) In [M(H2O)6][SiF6], H2O and [M(H 2O)6]2+ undergo 180° flips and reorientation about the C3, axis, respectively. The order-disorder transition is closely related to a freezing of this reorientation. (4) By elongation of [M(H2O)6]2+ along the C 3 axis, the mobility becomes higher. (5) The disorder of [Fe(H 2O)6]2+ in the high-temperature phase of [Fe(H2O)6][SiF6] is dynamic. Rotational modulation of [Mg(H2O)6]2+ along the C 3 axis exists in the incommensurate phase of [Mg(H2O) 6] [SiF6].

The possibility for proximity of CeRu2Si2 to the quantum critical point without any doping is discussed.

imuli including mechanical stress and plays important roles in pathophysiological conditions. For example, gravitational stress has been shown to induce expression of COX-2 in bone tissues, which is essential for bone homeostasis. To investigate whether COX-2 is induced by gravitational loading in other tissues than bone, we exposed mice to hypergravity at 2G and 3G for 4 h. We demonstrate here that COX-2 is induced in the mouse heart vessels by hypergravity. Moreover, hypoxia-inducible factor (HIF)-1α and its downstream genes such as inducible nitric oxide synthase, vascular endothelial growth factor, and heme oxygenase-1 were induced in the heart simultaneously, while none of these genes were induced in the COX-2(−/−) mouse heart. Therefore, COX-2 induced in the heart helps protect the heart function against hypoxia under hypergravity condition through HIF-1α induction.

atrix metalloproteinases (MMP) but also a cofactor for membrane-type 1 MMP (MT1-MMP)-mediated MMP-2 activation, it is still unclear how TIMP-2 regulates MMP-2 activation and cleavage of substrates by MT1-MMP. In the present study we examined the levels of cell-surface MT1-MMP, MMP-2 activation and cleavage of MT1-MMP substrates in 293T cells transfected with the MT1-MMP and TIMP-2 genes. Co-expression of TIMP-2 at an appropriate level increased the level of cell-surface MT1-MMP, both the TIMP-2-bound and free forms, and generated processed MMP-2 with gelatin-degrading activity. In contrast, MT1-MMP substrates testican-1 and syndecan-1 were cleaved by the cells expressing MT1-MMP, which was inhibited by TIMP-2 even at levels that stimulate MMP-2 activation. These results suggest that TIMP-2 environment determines MT1-MMP substrate choice between direct cleavage of its own substrates and MMP-2 activation. © 2007 Japanese Cancer Association.

ffraction. The crystal, C5H6N2O2S, belongs to space group P1 with cell dimensions of a = 4.9865(7)Å, b = 5.5716(7)Å, c = 12.544(2)Å, a = 74.793(8)°, b = 80.413(9)°, g = 85.001(10)°. The final R1 value is 0.036 for 1304 reflections (I > 2.0s(I)). In the new polymorph, intermolecular hydrogen bonds are formed between the acetyl C=O and the amide N-H groups [N(1)·O(2)(i) 2.800(2)Å, N(1)-H·O(2)(i) 151.02o, symmetry codes: (i) x+1, y-1, z] to form an infinite chain structure. On the other hand, the hydrogen bonds in the known form are formed between the amide C=O and the amide N-H groups of the thiohydantoin rings. 2009 © The Japan Society for Analytical Chemistry.

f nicotine addiction. Quantitative and qualitative differences in nicotine addiction have been observed between ethnic groups. However, there are few data on the ethnic influences of the CYP2A6-nicotine metabolism relationship, particularly with regard to black subjects. We determined the nicotine metabolism and CYP2A6 genotype in 176 white subjects and 160 black subjects, comparing them with our previous data from 209 Korean subjects and 92 Japanese subjects. Large interindividual differences were observed in the cotinine/nicotine ratios in plasma calculated as an index of nicotine metabolism in white subjects (range, 0.6-36.5) and in black subjects (range, 0.9-30.4). No ethnic difference in the metabolic ratio was observed among white subjects (mean, 7.2 ± 5.0), black subjects (mean, 7.1 ± 4.7), and Korean subjects (mean, 8.7 ± 11.9), whereas Japanese subjects showed a significantly (P < .005) lower metabolic ratio (mean, 3.8 ± 3.1) compared with the other populations. Women showed significantly (P < .05) higher metabolic ratios than men in the black population (8.0 ± 5.3 versus 6.0 ± 3.7). Obvious ethnic differences in the CYP2A6 alleles were observed among these 4 populations. The combined frequencies of the alleles lacking or showing reduced enzymatic activity (CYP2A6*2, CYP2A6*4, CYP2A6*5, CYP2A6*7, CYP2A6*9, CYP2A6*10, CYP2A6*11, CYP2A6*17, CYP2A6*19, and CYP2A6*20) were 9.1%, 21.9%, 42.9%, and 50.5% in white, black, Korean, and Japanese subjects, respectively. These CYP2A6 alleles were associated with reduced nicotine metabolism. Among the homozygotes of CYP2A6*1, interindividual and ethnic differences in the metabolic ratio were still observed. Thus some factors other than genetic ones might also contribute to the interindividual and ethnic differences. This comprehensive study of 4 populations extends our understanding of nicotine metabolism and the impact of genetic polymorphisms of the CYP2A6 gene. © 2006 American Society for Clinical Pharmacology and Therapeutics.

ne regeneration using 3 different implant materials in rabbit mandibles and to compare the bone regenerative capability of these materials in an animal model. Study design: Adult male Japanese white rabbits (n = 48; 12-16 wks old; 2.5-3.0 kg) were divided into 4 groups, consisting of 12 animals each. The implant materials were β-tricalcium phosphate (β-TCP), autologous bone derived from the radius, and recombinant human bone morphogenetic protein 2 (rhBMP-2) with polylactic acid/polyglycolic acid copolymer and gelatin sponge (PGS) complex. After incising along the inferior border of the mandible, the materials were implanted as only grafts and covered by titanium mesh with screws. No material was implanted into the control group. The rabbits were killed at 2, 4, 8, 12, and 24 wks postoperatively, and formalin-fixed specimens containing titanium mesh were embedded in acrylic resin. The specimens were stained with hematoxylin and eosin. For immunohistochemical analysis, the specimens were treated with BMP-2 and fibroblast growth factor 2 (FGF-2) antibodies. Finally, they were examined microscopically. Results: The autologous bone induced substantially more new bone formation compared with β-TCP at 4 wks postoperatively. However, rhBMP-2/PGS induced new bone formation at 8 wks postoperatively. No growth of bony tissue was observed in the control group at any period. In the autologous bone and rhBMP-2/PGS groups, both BMP-2 and FGF-2 were observed later in the β-TCP group than in other groups. Conclusion: This study suggests that autologous bone as well as rhBMP-2/PGS implants induce expression of both BMP-2 and FGF-2 specifically at the operated sites, even at early stages. © 2007 Mosby, Inc. one(I) has been determined by X-ray diffraction. The crystal belongs to space group C2/c with cell dimensions of a = 23.81(1)Å, b = 9.884(5)Å, c = 12.536(8)Å, β = 117.38(1)°. The compound, C18H14N4O2S2·2C2H6OS, crystallizes with half a molecule in the asymmetric unit, the molecule being centrosymmetric. The centre of inversion lies on the middle of the C(3)-C(3)′ bond. In crystals, the molecules form two N-H…O cyclic hydrogen bonds in a cyclic R22(12) arrangement, each of which forms on either side. DMSO molecules are hydrogen-bonded to the amide N-H groups. This hydrogen bond network forms an infinite tape structure along the c-axis.

We examined the expression of E2F1 and the aberration of RB gene and protein to elucidate what factors contribute to the overexpression of E2F1 in non-small cell lung carcinomas. Methods The expression level of E2F1 in tissues of non-small cell lung carcinomas was measured by means of quantitative reverse transcription-polymerase chain reaction and immunohistochemistry. For RB, we examined loss of heterozygosity (LOH) by PCR-restriction fragment length polymorphism and a variable number of tandem repeats, and protein expression by immunohistochemistry. Results Fifteen cases of carcinoma (46%) showed high transcription levels of E2F1 gene. Immunohistochemically, almost all (14 of 15) cases overexpressing E2F1 mRNA were positive for E2F1 protein. LOH at the RB locus was found in 13 of 30 informative cases. In 13 cases with LOH, ten showed overexpression of E2F1 mRNA and protein. Immunohistochemical positivity for phosphorylated RB protein was also closely correlated with overexpression of E2F1. Conclusions Our results suggest that overexpression of E2F1, induced both by LOH at the RB locus and anomalous phosphorylation of the RB protein, is involved in the development of non-small cell lung carcinoma.

ing sterically bulky substituent(s) on the terminal nitrogens has been investigated, where L = tris(N-benzylaminoethyl)amine (LH,Bn), tris(N-benzyl-N-methylaminoethyl)amine (LMe,Bn), or tris(N,N-dimethylaminoethyl)amine (LMe,Me). All the copper(I) complexes reacted with dioxygen at low temperatures to produce superoxocopper(II) and/or trans-(μ-1,2-peroxo)-dicopper(II) complexes depending on the steric bulkiness of the terminal nitrogens and the reaction conditions. The reaction of a copper(I) complex [Cu(LH,Bn)]+ at −90 °C in acetone resulted in the formation of a superoxo complex [Cu(LH,Bn)(O2)]+ as a less stable species and a peroxo complex [{Cu(LH,Bn)}2(O2)]2+ as a stable species. The structures of [Cu(LH,Bn)]ClO4 and [{Cu(LH,Bn)}2(O2)](BPh4)2·8(CH3)2CO were determined by X-ray crystallography. [{Cu(LH,Bn)}2(O2)]2+ has a trans-(μ-1,2-peroxo)-dicopper(II) core with a trigonal bipyramidal structure. The O–O bond distance is 1.450(5) Å with an intermetallic Cu···Cu separation of 4.476(2) Å. The resonance Raman spectrum of [{Cu(LH,Bn)}2(O2)]2+ measured at −90 °C in acetone-d6 showed a broad ν(O–O) band at 837–834 cm−1 (788 cm−1 for an 18O labeled sample) and two ν(Cu–O) bands at 556 and 539 cm−1, suggesting the presence of two peroxo species in solution. [Cu(LMe,Bn)]+ also produced both superoxo and trans-μ-1,2-peroxo species, [Cu(LMe,Bn)(O2)]+ and [{Cu(LMe,Bn)}2(O2)]2+. At a lower concentration of [Cu(LMe,Bn)]+ (∼0.24 mM) and higher dioxygen concentration (P(O2) = ∼1 atm), the superoxo species is predominantly formed, whereas at a higher concentration of [Cu(LMe,Bn)]+ (∼1 mM) and lower dioxygen concentration (P(O2) = ∼0.02 atm) the formation of the peroxo species is observed. The resonance Raman spectrum of [Cu(LMe,Bn)(O2)]+ (∼1 mM) in acetone-d6 at ∼−95 °C exhibited a ν(O–O) band at 1120 cm−1 (1059 cm−1 for an 18O labeled sample) and that of [{Cu(LMe,Bn)}2(O2)]2+ (∼3 mM) in acetone-d6 at ∼−90 °C showed two ν(O–O) bands at 812 and 797 cm−1 (767 and 753 cm−1 for an 18O labeled sample), respectively. A similar observation was also made for [{Cu(LMe,Me)}2(O2)]2+. Relationships between the energies of the LMCT and d–d transitions and those of the ν(O–O) and ν(Cu–O) stretching vibrations and the steric constraints in the Cu(II)–(O22−)–Cu(II) core are discussed.

disubstituted 2,2′-bipyridine (Y2bpy; Y = H (bpy), Me, Cl, N(Et)2, CONH2 or COOEt) or 2,2′-bipyrimidine) and an aromatic amino acid (AA = l-phenylalanine (Phe), p-substituted phenylalanine (XPhe; X = NH2, NO2, F, Cl or Br), l-tyrosine (Tyr), l-tryptophan (Trp) or l-alanine (Ala)) were characterized by X-ray diffraction, spectroscopic and potentiometric measurements. The structures of [Cu(dpa)(Trp)]ClO4·2H 2O and [Cu((CONH2)2bpy)(Phe)]ClO 4·H2O in the solid state were revealed to have intramolecular π-π interactions between the Cu(ii)-coordinated aromatic ring moiety, Cu(DA) (Mπ), and the side chain aromatic ring of the AA (Lπ). The intensities of Mπ-Lπ interactions were evaluated by the stability constants of the ternary Cu(ii) complexes determined at 25 °C and I = 0.1 M (KNO3), which revealed that the stability enhancement of the Cu(DA)(AA) systems due to the interactions is in the order (CONH 2)2bpy < bpy < Me2bpy < (Et 2N)2bpy with respect to DA. The results indicate that the electron density of coordinated aromatic diimines influences the intensities of the stacking interactions in the Cu(DA)(AA) systems. The Mπ-Lπ interactions are also influenced by the substituents, X, of Lπ and are in linear relationship with their Hammett σp values with the exception of X = Cl and Br. © The Royal Society of Chemistry

, which is a homologue of transient receptor potential (TRP) cation channel proteins. Here we identify a novel PKD2 mutation that generates a C-terminal tail-truncated TRPP2 mutant 697fsX with a frameshift resulting in an aberrant 17-amino acid addition after glutamic acid residue 697 from a family showing mild ADPKD symptoms. When recombinantly expressed in HEK293 cells, wild-type (WT) TRPP2 localized at the endoplasmic reticulum (ER) membrane significantly enhanced Ca2+ release from the ER upon muscarinic acetylcholine receptor (mAChR) stimulation. In contrast, 697fsX, which showed a predominant plasma membrane localization characteristic of TRPP2 mutants with C terminus deletion, prominently increased mAChR-activated influx in cells expressing TRPC3 or TRPC7. Coimmunoprecipitation, pulldown assay, and cross-linking experiments revealed a physical association between 697fsX and TRPC3 or TRPC7. 697fsX but not WT TRPP2 elicited a depolarizing shift of reversal potentials and an enhancement of single-channel conductance indicative of altered ion-permeating pore properties of mAChR-activated currents. Importantly, in kidney epithelial LLC-PK1 cells the recombinant 679fsX construct was codistributed with native TRPC3 proteins at the apical membrane area, but the WT construct was distributed in the basolateral membrane and adjacent intracellular areas. Our results suggest that heteromeric cation channels comprised of the TRPP2 mutant and the TRPC3 or TRPC7 protein induce enhanced receptor-activated Ca2+ influx that may lead to dysregulated cell growth in ADPKD. © 2009 by The American Society for Biochemistry and Molecular Biology, Inc.<br />Publisher's version/PDF may be used after 12 months embargo

In the 5-BTH crystals, the enantiomeric (R)- and (S)-5-BTH molecules are connected to form cyclic dimers via the hydrogen bonds of the thioamide and the amide moieties. On the other hand, the intermolecular hydrogen bonds in 1-Ac-5-BTH crystals form an infinite chain. These differences in the hydrogen bond pattern are also discussed in the IR and Raman spectra. The ab initio molecular orbital calculations (Gaussian 03) with 6-31G(d,p) basis set were carried out for 5-BTH and 1-Ac-5-BTH to get the preferred conformation. The molecules differ mainly in the orientation of the phenyl ring with respect to the five-membered ring. Changes in the enantiomeric excesses (ee) of chiral substances with circularly polarized light (CPL) irradiation have been simulated, and the necessary conditions for observing the MFE were obtained. The asymmetric photolysis of racemic 2-phenylcycloalkanones (2-PCAs) with CPL has been carried out in both the presence and absence of a magnetic field. Since the anisotropy g factors of 2-PCAs are considerably large, the CPL-induced ee are achieved to a few percent after 90% decomposition though the MFE are not observed. The photolysis mechanism of 2-PCAs in an air-saturated solution has been also clarified. Triplet acyl–benzyl biradicals, formed via photochemical α-cleavage of 2-PCAs, react with O2 dissolved in the solution and result in the formation of acetophenone and alkenoic acids. The bimolecular reactions are diffusion-controlled and the rates are comparable to those of ISC to the singlet biradicals for all 2-PCAs. The recombination yields of the biradicals are sufficiently large. However, the biradical ISC rate shows little magnetic field dependence, which explains the absence of the MFE in this asymmetric photolysis.

prostaglandin I2 (PGI2) remains obscure. This study was designed to investigate the role of PGI2 in cough reflex sensitivity of the asthmatic airway, which is characterized by chronic eosinophilic airway inflammation. The effect of beraprost, a chemically and biologically stable analogue of PGI2, on cough response to inhaled capsaicin was examined in 21 patients with stable asthma in a randomized, placebo-controlled cross over study. Capsaicin cough threshold, defined as the lowest concentration of capsaicin eliciting five or more coughs, was measured as an index of airway cough reflex sensitivity. The cough threshold was significantly (p < 0.05) decreased after two weeks of treatment with beraprost [17.8 (GSEM 1.20) μM] compared with placebo [30.3 (GSEM 1.21) μM]. PGI2 increases cough reflex sensitivity of the asthmatic airway, suggesting that inhibition of PGI2 may be a novel therapeutic option for patients with asthma, especially cough predominant asthma.

The change of [Ca2+]i was mediated by muscarinic type 3 (m3) AChR. Both phosphatidylinositol-specific phospholipase C blocker (U73122) and IP3R blocker (2-APB) inhibited ACh-induced [Ca2+]i elevation, suggesting that IP3 pathway is involved in [Ca2+]i alteration. Ryanodine and FK506 increased ACh-induced [Ca2+]i elevation, which was decreased by RyR blocker (ruthenium red). These results suggest that RyR and FK506 binding protein 12 kDa (FKBP12) complex was involved in [Ca2+]i change, and that TE671/RD cell line has a hybrid characteristic of smooth and skeletal muscles. the cyclocondensations with oxalyl chloride in the presence of tricthylamine at 0°C has been shown analogous to that obtained for 1,2-disubstituted acyclic ethylene glycols 1: cis-1,2-cyclooctanediol (9f) produced the cyclic oxalate 14f as the major product, while trans-1,2-cycloheptanediol (9e) and trans-1,2-cyclooctanediol (9g) formed the cyclic carbonates 12e, g as the major products. On the other hand, the cyclic oxalates 14a-d were formed as the major products from 1,2-cyclopentane- and 1,2-cyclohexanediols regardless of the configuration. These results can be accounted for by assuming the boat-like transition states for cyclizations of the half esters of comparatively rigid five- and six-membered diols 9a-d. The cyclic oxalates 14a, c may be directly formed through the resulting tetrahedral intermediates from cis-diols (9a, c), and the cyclic carbonates 12a, c as the minor products after ring inversion of the tetrahedral intermediates. The tetrahedral intermediates from the trans-isomers 9b,d cannot undergo ring inversion, producing no traces of the cyclic carbonates 12b,d. © 2002 Pharmaceutical Society of Japan.

on after implantation of a statin and recombinant human BMP-2 (rhBMP-2) and to compare the bone regeneration capability of these substances in the rabbit nasal bone using immunohistologic methods. Study design: Twelve adult male Japanese white rabbits (n = 12; age 12-16 weeks, weight 2.5-3.0 kg) were divided into 3 experimental groups and 1 control group. A total of 48 bone defects, 4 per rabbit, were created in the nasal bone while preserving the nasal membrane. In the experimental groups, 1 group was implanted with 10 mg of a statin dissolved in 0.2 mL water with an atelocollagen sponge (ACS); the second group was implanted with 5 μg rhBMP-2 with an ACS; and in the third group only the ACS was implanted. No material was implanted in the control group. Animals were killed at 1, 2, and 4 weeks after surgery. The parts that had been operated on were removed and prepared for histologic assessment. The expression of BMP-2 was evaluated using immunohistochemistry, and double-immunostaining for BMP-2 and Ki-67 was observed by fluorescent microscopy. Results: No significant differences were observed between the statin/ACS group and rhBMP-2/ACS group at 1, 2, and 4 weeks after surgery. The number of cells which stained positively for BMP-2 increased significantly in both of the implanted groups compared with the control group (P < .0001). The positive fluorescent double-immunostaining for BMP-2 and Ki-67 was similar in both implanted groups. Conclusion: This study suggests that statin/ACS implants show BMP-2 expression and osteoinductive activity that is similar to those of rhBMP-2/ACS implants. © 2009 Mosby, Inc. All rights reserved..

am product of cyclooxygenase 2 (COX-2), plays a key role in gastric tumorigenesis. The Wnt pathway is also suggested to play a causal role in gastric carcinogenesis. However, the molecular mechanism remains poorly understood of how the Wnt and PGE2 pathways contribute to gastric tumorigenesis. To investigate the role of Wnt and PGE2 in gastric cancer, we have generated transgenic mice that activate both pathways and examined their phenotypes. Methods: We constructed K19-Wnt1 transgenic mice expressing Wnt1 in the gastric mucosa using the keratin 19 promoter. We then crossed K19-Wnt1 mice with another transgenic line, K19-C2mE, to obtain K19-Wnt1/C2mE compound transgenic mice. The K19-C2mE mice express COX-2 and microsomal prostaglandin E synthase-1 (mPGES-1) in the stomach, showing an increased gastric PGE2 level. We examined the gastric phenotypes of both K19-Wnt1 and K19-Wnt1/C2mE mice. Results: K19-Wnt1 mice had a significant suppression of epithelial differentiation and developed small preneoplastic lesions consisting of undifferentiated epithelial cells with macrophage accumulation. Importantly, additional expression of COX-2 and mPGES-1 converted the preneoplastic lesions in the K19-Wnt1 mice into dysplastic gastric tumors by 20 weeks of age. Notably, we found mucous cell metaplasia in the glandular stomach of the K19-Wnt1/C2mE mice as early as 5 weeks of age, before the dysplastic tumor development. Conclusions: Wnt signaling keeps the gastric progenitor cells undifferentiated. Simultaneous activation of both Wnt and PGE2 pathways causes dysplastic gastric tumors through the metaplasia-carcinoma sequence. © 2006 American Gastroenterological Association (AGA) Institute.

TGF-β) plays a pivotal role in tissue repair and fibrosis. The early growth response-1 (Egr-1) transcription factor mediates profibrotic TGF-β responses, and its expression is elevated in biopsies from patients with scleroderma. NGF1-A-binding protein 2 (Nab2) is a conserved transcriptional cofactor that directly binds to Egr-1 and positively or negatively modulates Egr-1 target gene transcription. Despite the recognized importance of Nab2 in governing the intensity of Egr-1-dependent responses, the regulation and function of Nab2 in the context of fibrotic TGF-β signaling is unknown. Here we show that TGF-β caused a time-dependent stimulation of Nab2 protein and mRNA in normal fibroblasts. Ectopic expression of Nab2 in these cells blocked Egr-1-dependent transcriptional responses, and abrogated TGF-β-induced stimulation of collagen synthesis and myofibroblasts differentiation. These inhibitory effects of Nab2 involved recruitment of the NuRD chromatin remodeling complex to the COL1A2 promoter and were accompanied by reduced histone H4 acetylation. Mice with targeted deletion of Nab2 displayed increased collagen accumulation in the dermis, and genetic or siRNA-mediated loss of Nab2 in fibroblasts was associated with constitutively elevated collagen synthesis and accentuation of Egr-1-dependent TGF-β responses in vitro. Expression of Nab2 was markedly up-regulated in skin biopsies from patients with scleroderma, and was localized primarily to epidermal keratinocytes. In contrast, little Nab2 could be detected in dermal fibroblasts. These results identify Nab2 as a novel endogenous negative regulator of Egr-1-dependent TGF-β signaling responsible for setting the intensity of fibrotic responses. Defective Nab2 expression or function in dermal fibroblasts might play a role in persistent fibrotic responses in scleroderma. © 2008 Bhattacharyya et al.

ysfunction of pancreatic β-cells. Furthermore, the number of pancreatic β-cells decreases as a secondary effect of advanced type 2 diabetes, although the molecular mechanism has not been elucidated. Recently, it has been shown that hyperglycemic conditions induce the expression of cyclooxygenase-2 in pancreatic islets and increase the downstream product prostaglandin E2 (PGE2). To investigate whether high glucose-induced PGE2 has an adverse effect on pancreatic β-cells, we generated transgenic mice (RIP-C2mE) that express cyclooxygenase-2 and microsomal prostaglandin E synthase-1 in their β-cells using the rat insulin-2 gene promoter (RIP). The homozygous RIP-C2mE (Tg/Tg) mice showed severe hyperglycemia from six weeks of age. Although the heterozygous RIP-C2mE (Tg/-) mice showed normal blood glucose levels throughout their lifetime, this level increased significantly compared with that of wild-type mice when glucose was loaded. The relative number of β-cells to the total islet cell number was reduced to 54 and 14% in the RIP-C2mE (Tg/-) and (Tg/Tg) mice, respectively, whereas that in the wild-type mice was 84%. Importantly, the proliferation rate in the islets of the RIP-C2mE (Tg/Tg) mice at four weeks of age decreased significantly in comparison to that in the wild-type mice. Because β-cells replicate not only during the postnatal period but also in the adult pancreas at a basal level, it is possible that increased PGE2 signaling thus contributes to the reduction of the pancreatic β-cell mass through inhibition of proliferation, thereby aggravating diabetes further. © 2006 by The American Society for Biochemistry and Molecular Biology, Inc.

We analyzed the aberrant methylation of TFPI-2 in the pure pancreatic juice (PPJ) aspirated endoscopically from patients with various pancreatic diseases. Using the highly sensitive methylation-specific polymerase chain reaction (MSP) and quantitative MSP (Q-MSP) assay, we investigated the aberrant methylation of TFPI-2 in nine human PCa cell lines and in the PPJ from patients with PCa, intraductal papillary mucinous neoplasms (IPMN) and chronic pancreatitis (CP). The incidence of aberrant TFPI-2 methylation was seven (77.8%) of nine PCa cell lines by Q-MSP. In cell lines, the expression of TFPI-2 mRNA by quantitative reverse transcription-polymerase chain reaction showed an inverse correlation to the aberrant methylation of TFPI-2. The incidence of aberrant TFPI-2 methylation in the PPJ was 21 (58.3%) of 36 PCa patients, three (17.6%) of 17 IPMN and one (4.8%) of 21 CP by MSP assay. Using a suitable cut-off value of 2.5 according to the receiver operating characteristic curve, the incidence of aberrant TFPI-2 methylation in the PPJ by real-time MSP was 18 (62.1%) of 29 PCa patients, one (5.1%) of 17 IPMN and three (14.3%) of 21 CP, respectively. The incidence of quantitative TFPI-2 hypermethylation in the PPJ with PCa was significantly higher than that with IPMN (P < 0.001) or CP (P < 0.001). Moreover, the aberrant methylation rate of TFPI-2 in the PPJ was 100%, as observed (6/6) in the PCa patients with liver metastasis, and 86.7% (26/30) in stages IVa + IVb of PCa by Q-MSP assay. These results suggest that promoter methylation of TFPI-2 in the PPJ may be a useful marker in the diagnosis and progression of PCa using an endoscopically feasible approach. © 2006 Japanese Cancer Association.

The abnormal UV-vis absorption spectrum in an acidic solution can be explained by the existence of the bulky and electronegative CF3 group at the 2-position, which prevents protonation at the Nβ atom of the azo group in an acidic solution.

S2- molecules in the vacuum and found that the calculated equilibrium bond lengths and vibrational spectroscopic constants are in good agreement with the experiments as well as the previous calculations. Based on this conformation we extended these calculations to the substitutional S2- molecule in the alkali halides, such as NaCl, NaBr, NaI, KCI, KBr, KI, RbCl, RbBr, and RbI. It is found from these calculations that the S2- center aligned to [110] direction is generally favorable over those with the [100] and [111] orientations, in agreement with the experiments. We also found that the vibrational frequencies of the S2- anion in the alkali halides shift to higher energies with decreasing lattice parameters. These results suggest that the change in the bond length of the S2- anion in the matrices depends on the species of halogen atom, while the frequencies are influenced by the species of alkali atom.
