目し、感覚情報入力と行動出力の対応関係が明確な性フェロモン伝達神経回路の可視化と機能解析を目的とした。本研究では、本研究者が発見した神経活動依存的な発現増加を示す遺伝子Hr38に着目し、カイコガとショウジョウバエの2種の昆虫を用いて、活動依存的な神経回路可視化法及び機能解析法を構築した。本手法を用いることで、既知の回路に加え新規な神経回路が性フェロモン情報処理に関与することを見出した。<br />How do animals select their behavior in response to certain sensory inputs? Appropriate response to sensory information is critical for animal survival. In the present study, I focused on the brain of insects which has low complexity, but has similar circuit property and molecular basis as the brain of other animals including vertebrates. In many insects, sex pheromone induces stereotypic courtship behavior with clear relationship between receptor and behavior. Here, I established a novel neural circuit tracing method utilizing an activity-dependent gene expression. Recently, I identified a novel immediate early gene Hr38, whose expression increases in response to neural activity. In the present study, I used the promoter activity of Hr38 to express GFP in order to visualize neurons activated by sex pheromone or courtship behavior. 神経活動依存的に発現量が増加する新規な遺伝子としてHr38を同定し、これがカイコガ・ショウジョウバエの脳において神経活動のマーカーとして利用できることを見出し、異性の匂いに応答する細胞の包括的同定に成功した。さらに遺伝子組換え昆虫を用いて神経活動の起こった神経細胞特異的にGFPなどのレポーター遺伝子を発現する昆虫の作成を試みた。<br />Many insects exhibit stereotypic instinctive behavior, but the underlying neural mechanisms are not well understood due to difficulties in detecting brain activity in freely-moving animals. Immediate early genes (IEGs) whose expression is transiently and rapidly upregulated upon neural activity are powerful tools for detecting behavior-related neural activity. In the present study, I identified Hr38 as a novel IEG that is transiently expressed in the male silkmoth, Bombyx mori, by female odor stimulation. Using Hr38 expression as an indicator of neural activity, I mapped comprehensive activity patterns of the silkmoth brain in response to female sex pheromones. I found that Hr38 can also be used as a neural activity marker in the fly, Drosophila melanogaster. Using Hr38, I constructed a neural activity map of the fly brain in response to female stimulation.
