概要: 金沢大学医薬保健研究域医学系<br />体内時計は糖・脂質代謝など様々な生理機能の恒常性維持に重要な役割を果たしていることが、臓器特異的時計遺伝子欠損マウスの解析などにより示唆されている。しかし、正常に機能する体内時計が障害された場合の影響 、特に肝や脂肪組織などの末梢時計障害の影響は、十分に解明されていない。本研究では、正常マウスを用い、照明条件あるいは給餌時刻を変更することにより、全身性および末梢特異的体内時計障害モデルをそれぞれ開発した。両モデルとも摂餌量は低下するにもかかわらず体重への影響は認めず、前者では耐糖能の悪化も認めた。このことより、中枢時計と末梢時計はどちらも代謝の調節に重要な役割を果たしていることが示された。<br />The results of studies using tissue-specific clock gene knock out mice etc. suggest that circadian clocks play a significant role in maintaining homeostasis of various physiological functions including glucose and lipid metabolisms. However, it remains to be seen what happens if circadian clocks (in particular, peripheral clocks) are disrupted in bodies with innately intact clocks. In this study, we've developed the models of systemic and peripheral tissues-specific disruption of circadian clocks by changing the light/dark cycle or feeding time, respectively, using normal mice. Consequently, decreased food intake without change in body weight was found in both the models, and impaired glucose tolerance was seen in the systemic model. 概要: 金沢大学医薬保健研究域医学系 / 自治医科大学<br />肝や脂肪組織などの末梢組織における体内時計(末梢時計)の障害は、2型糖尿病を初めとする生活習慣病の一因であることが示唆されている。そこで、末梢時計を標的とした生活習慣病の予防・治療法 を開発するために、末梢時計の制御が可能な薬物をマウスモデルを用いて探索し、2型糖尿病治療薬であるエキセナチドには摂餌刺激よりも強力な末梢時計制御作用があることを見出した。また、空腹時血糖の日内リズムは直接的には肝の体内時計により制御されていることを明らかにし、糖代謝制御機構における末梢時計の関与を明確にした。<br />Recent studies have indicated that disrupted circadian clocks in peripheral tissues (peripheral clocks) are associated with the pathophysiology of lifestyle-related metabolic diseases including type 2 diabetes. To develop a strategy targeting peripheral clocks for treatment/prevention of metabolic diseases, whether exenatide could affect the rhythms of peripheral clocks was investigated in mice, and we found that exenatide modulates peripheral clocks more strongly than feeding. 概要: 金沢大学医薬保健研究域医学系<br />褐色脂肪細胞(BA)の体内時計の病態生理学的意義を明らかにするために、BA特異的に体内時計を欠損したマウスを作製した。このマウスでは、BAにおいて脂肪酸を利用した熱産生の障害が認められ、自発運動量の増 加や骨格筋のふるえによる熱産生を増加させて代償的に体温を維持していた。また、エネルギー消費量が少なく、高脂肪食負荷時には肥満が増悪した。したがって、BAの体内時計は熱産生の制御に重要であり、肥満・生活習慣病の予防・治療標的となることが示唆された。<br />To elucidate the pathophysiological role of circadian clock in brown adipocytes (BA), BA-specific clock knockout mice were generated. This strain showed impairment of thermogenesis using fatty acids and maintained body temperature by increasing shivering and behavioral thermogenesis. Moreover, the conditional knockout mice exhibited reduced energy expenditure and were more prone to diet-induced obesity.
